Not sure what you mean.
If basic, usually things like adjusting your seat, leg distance, your mirrors etc.....

Yep, make sure

1) Car got petrol
2) Tyres are okay, have tread, properly inflated
3) Sufficient Fluids, water, coolant, brake, engine oil
4) Lights and Wipers Working
5) Air-cond working

Lastly, driver has enough rest...

Say, you're preparing for a cross-country expedition driving from KL to Lhasa and back, a trip of 50 days.

What then would be your preparation?

Make sure your car battery is functioning properly and has enough battery water to sustain its life. You should always take a good health check up of the car without fail before starting for a drive as there should be no problem when you are on the move.
